(AP) ? We've all been there before.

Walking down the street or shopping for a shirt when we find ourselves chest-to-belt buckle with some impossibly tall, muscled platinum member of the human race whose inescapable physical perfection reminds us that it's been a few days ? er, weeks ? uh, months ? since we hit the gym.

At glittering Westfield Mall on the edge of Olympic Park, uncomfortable encounters between muscle and mush are remarkably frequent. At least here the Adonises wear team uniforms, so there's no mistaking them for regular people who are simply much more fit than you.

These are Olympians. No wonder they look better than the rest of us.

Still, it's hard to hold back feelings of inferiority, whether you are a girthful Londoner, a pasty journalist or one of the multitudes of candy apple-shaped tourists who have come from around the world to witness the games.

Flabbiness knows no borders.

"It's intimidating," said Richard Perry, a Londoner buying a soft, sugared concoction at the mall's Mr. Pretzel concession stand Saturday as two mammoth Australian Olympians strutted past. "It's like they are from a different planet."

Over there are some telegenic water polo players buying coffee and posing with fans. And wait a second: Isn't that chiseled Belgian judo fighter Daniel Fernandes, chatting with a portly security guard? And here comes a 6-foot-4-inch Russian woman in a national team tracksuit, window-shopping but refusing questions with a withering "nyet."

Hopefully, when more competitions get under way in the coming days, these Masters of the Universe will be too busy to shop, and will remain locked away in the athletes village, where mortals are not allowed to tread and we are protected from their awesomeness by a tall metal fence.

It's not that we resent them. After all, we are here to celebrate them.

But it's one thing to watch them throw a javelin and entirely another to stand waiting behind their hulking presence to buy a pair of sunglasses, nervously pretending to read something on your smartphone.

Indeed, the superhuman bodies of many of the men and women competing in the games are a stark reminder of how far the rest of us have fallen.

The host country may be expecting a record haul of gold medals from its world-class athletes, but its people really pack on the British pounds. This is the fattest country in Europe, with a quarter of the population officially obese.

A 2009 study found one in six Britons were too lazy to get up to change the channel if they couldn't find the remote, and would instead continue watching a program they were not interested in.

Americans are even more overweight: More than a third of adults are obese, according to the Centers for Disease Control and Prevention. And they are sedentary. The average American logs four hours a day in front of the television, not to mention time spent online or texting friends.

Perry, the pretzel purchaser, said he was cautiously optimistic that the Olympic Games would inspire him to shed his modest middle-aged paunch. Others were unmoved, despite the example set by the world's finest athletes.

"They look great," gushed Lucy Harley, a young Apple Store employee at the mall who insisted she was comfortable with her average level of fitness ? and realistic about her laziness.

"If I wanted to look like that, I would dedicate more time to sport," she shrugged, making it clear she wasn't about to.

Elsewhere at the $2.75 billion shopper's paradise, ticket-holders gorged themselves on dark-chocolate ice-cream pops and rich cupcakes as big as a basketball player's fist. Belt-busting English beer flowed. American-style pizza was served in all its gooey greatness.

The throngs of diners at the spacious ground-floor McDonald's were at least as large and enthusiastic as the crowds that turned out to watch an Olympic cycling race through the heart of London.

Some athletes who were window-shopping as they waited for events to begin were happy to share advice ? and words of comfort ? with the mushy multitudes.

"In some sports, like weightlifting, we even have fat athletes," offered Marouane M'rabet, a lean Tunisian volleyballer who was sampling some massage cream that a vivacious sales clerk was enthusiastically rubbing onto his forearm.

He said anybody who wanted to get in shape should work out for half an hour a day, and insisted they would notice a difference in their physique very quickly.

Just don't expect to look like an Olympian.

"For an athlete? No. Thirty minutes is not enough," he laughed. "This is nothing."

HBT: Sanchez shelled in first game as Tigers pitcher

Anibal Sanchez stumbled in his Tigers? debut this afternoon, giving up five runs over six-plus innings in a 5-1 loss to the Blue Jays.

Sanchez, who was acquired from the Marlins this week along with second baseman Omar Infante, allowed eight hits and three walks. This included a season-high three home runs. Edwin Encarnacion and Yunel Escobar both had solo shots while Colby Rasmus? two-run blast in the bottom of the seventh chased Sanchez from the ballgame.

Sanchez?s season ERA jumped from 3.94 to 4.11 in the loss. The impending free agent has a 5.81 ERA over his last 10 starts dating back to the start of June. It will be interesting to see how he fares pitching in the more difficult league, but if healthy, his peripherals and track record both suggest that he should be a fine addition to the Tigers? rotation down the stretch.

Henderson Alvarez limited the Tigers to five hits over seven innings of one-run ball en route to improving to an even 7-7 on the year. The Blue Jays have won three in a row to move within 3 1/2 games in the Wild Card race while the Tigers have dropped three straight to fall two games behind the White Sox in the American League Central.

A look at recent tech-industry earnings

Samsung reported strong results Friday, days after rival Apple showed a slowdown in revenue growth. Here is a summary of recent earnings and reports for selected technology companies and what they reveal about the state of spending and the overall economy:

? July 2: Microsoft Corp. says it is absorbing a $6.2 billion charge to reflect that one of the biggest deals in its 37-year history, for online ad company aQuantive, turned out to be a dud.

? July 6: Samsung Electronics Co. estimates its second-quarter operating profit at 6.5 trillion won to 6.9 trillion won ($5.7 billion to $6.1 billion), a 79 percent jump from a year earlier based on the midpoint of that range. Analysts say the sharp rise in operating profit was driven by the success of its Galaxy line of smartphones. However, Samsung's stock fell as lower-than-expected overall sales underlined the threat from Europe's economic malaise.

? July 9: Chipmaker Advanced Micro Devices Inc. lowers its quarterly forecast, blaming weaker-than-expected sales in China and Europe and lackluster demand overall from consumers. AMD says it expects revenue to fall by 11 percent from the previous quarter, below its earlier forecast of a 3 percent decline.

? July 10: BlackBerry maker Research in Motion Ltd. asks disgruntled investors at a shareholders meeting for patience as it develops new devices to rival the iPhone and Android smartphones. The company reiterates a warning that the next several quarters will be challenging. It also expects to book another operating loss in the current quarter as the company cuts prices to sell its older BlackBerry models.

? July 17: Yahoo Inc. reports another lackluster quarter on CEO Marissa Mayer's first day on the job. It continues years of financial lethargy that has plagued the one-time Internet pioneer as Google and Facebook have prospered. Net income fell 4 percent from a year ago, and adjusted earnings were short of expectations.

Intel Corp., the world's largest chipmaker, says revenue for the current quarter is likely to come in below Wall Street forecasts as it blames "a more challenging macroeconomic environment."

? July 18: IBM Corp. reports its 38th consecutive quarter of earnings increase. Net income rose 6 percent despite a 3 percent drop in revenue amid Europe's economic jitters. IBM revenue in Europe, the Middle East and Africa fell by 9 percent from last year. The turmoil in Europe hurt IBM's results as currencies there weakened, translating into fewer U.S. dollars. IBM says revenue would have been slightly higher than last year if currency-exchange rates had remained the same.

EBay Inc. more than doubled its second-quarter net income thanks to higher revenue from its PayPal online payments business and its e-commerce websites. Its marketplaces business, which includes its flagship website, saw the strongest growth rate since 2006.

? July 19: Google Inc. says its second-quarter earnings rose 11 percent despite a deepening decline in the prices paid for its Internet search advertising. The results included Google's $12.5 billion acquisition of cellphone maker Motorola Mobility Holdings Inc. for the first time. Revenue climbed 35 percent from last year to $12.2 billion. If not for Motorola, revenue would have increased by 21 percent.

Microsoft Corp. says a $6.2 billion accounting adjustment to reflect a weak online ad business led to its first quarterly loss in its 26 years as a public company. The company had a loss of 6 cents a share, compared with earnings of 69 cents a share a year ago. Adjusted earnings beat expectations, and the stock rose.

Verizon Communications Inc. says net income rose 13 percent in the second quarter as its wireless arm pulled in record profits. But its traditional, wired phone business was weaker. Like other phone companies, Verizon is losing landlines, but has been compensating to some extent by signing up broadband customers. In the second quarter, that trend faltered, as it gained just 2,000 broadband customers ? the worst result in four years.

? Tuesday: Apple Inc. reveals that its growth slowed in the most recent quarter. In both revenue and net income, the company posted the smallest increases in years and failed to meet analyst expectations. It wasn't so much the volume of sales, but Apple's average selling prices declined.

At AT&T Inc., meanwhile, declining smartphone sales contributed to the best profitability ever in its wireless arm as it saved on phone subsidies. AT&T says it activated fewer smartphones in the quarter, much of it because subscribers are holding to their phones longer. That's good news for the company because it doesn't have to shell out as much in subsidies to sell the phones to customers for $99 or $199.

Netflix Inc. reports a 91 percent reduction in second-quarter earnings amid a slowdown in subscriber growth. Netflix Inc. added 1.1 million worldwide subscribers to its streaming service ? in line with management predictions, but below what analysts had hoped for.

? Wednesday: Online game maker Zynga Inc. reports a loss in the second quarter, with adjusted earnings and revenue below Wall Street's already low expectations. Zynga also lowers its outlook for the year, citing game delays, reduced expectations for "Draw Something" and what it called a "more challenging environment on the Facebook Web platform." Changes Facebook made to its website can make older games such as "FarmVille" more difficult to discover.

? Thursday: Facebook Inc.'s first earnings report as a public company had solid numbers, but in the end it landed with a thud ? much like its rocky initial public offering two months earlier. Facebook reported stronger-than-expected revenue and a gain in user numbers, but investors weren't impressed. After a brief spike, its stock fell.

Sprint Nextel Corp., the country's third-largest wireless carrier, reports a wider loss for the second quarter as wrote down the value of its moribund Nextel network. However, Sprint was successful in persuading smartphone subscribers to pay up for "unlimited data" service, and its service revenue zoomed, beating estimates.

Online retailer Amazon.com Inc. reports earnings that matched Wall Street's meager expectations, but its revenue gains and outlook for the current quarter fell short of forecasts.

? Friday: Samsung Electronics Co. reports another record-high quarterly profit as customers flocked to Galaxy smartphones. Samsung benefited from runaway demand for its Android-powered smartphones as rivals including Apple Inc. had yet to release new models. Robust sales of smartphones helped offset a slowdown in other consumer electronics businesses such as televisions.

Heterosexual men tend to go through HIV alone

TORONTO, July 27,2012 Heterosexual men make up a small but growing number of people infected with HIV in Canada. Yet a new study has found that many of them feel existing HIV-related programs and services don't meet their needs and are geared primarily or exclusively toward gay men and heterosexual women who are living with the virus.

"Heterosexual men tend to go through HIV alone," said Tony Antoniou, a pharmacist and research scholar in the Department of Family Medicine at St. Michael's Hospital. "They feel very isolated."

Antoniou said the study, published in the online medical journal BMJ Open, was among the first to examine how heterosexual men with HIV seek health care and support services.

While unprotected sex between men is still the most common method of transmitting HIV in North America and Western Europe, the number of new infections attributable to heterosexual transmission has been steadily increasing. Heterosexual men accounted for less than 5 per cent of all cases of HIV in Ontario before 1990, but represented 13.8 per cent of HIV diagnoses among men in 2008. In Britain, the figure is even higher -- 18.8 per cent of patients accessing HIV-related care in 2009 were men infected via heterosexual sex

Antoniou and his colleagues conducted focus groups of HIV-positive heterosexual men with the assistance of community-based AIDS service organizations in Ontario.

The men told them:

  • They perceive themselves to be relegated to the margins of a health care and service ?eld that was developed to address the needs of groups who were historically affected most by the illness namely, gay men and heterosexual women
  • They felt that gay men were better positioned to obtain and advocate for HIV-related services and support because of their "social capital" within AIDS service organizations
  • Many were reluctant to seek treatment or support or even be seen entering AIDS clinics because of ongoing societal stigma associated with HIV. In particular, they were concerned that their jobs could be in jeopardy if their HIV status were known or that their children would be stigmatized and shunned socially

"Heterosexual men living with AIDS are navigating a health care and service field that has not kept pace with their increased representation among the population of people living with the virus," Antoniou wrote in his study, Waiting at the dinner table for scraps: a qualitative study of the help-seeking experiences of heterosexual men living with HIV infection.

"Nobody is looking at them as husbands and fathers, people who need to be supported in those roles," he said. "In keeping with the tradition of involving people living with HIV in developing services to meet their concerns, researchers and policy makers should strive to integrate heterosexual men living with HIV in decision-making and community-based research initiatives."

TJ Jackson Pushing for Guardianship of Children in Face of Alleged Katherine Jackson Kidnapping


The Jackson family saga is growing more sad, disturbing and confounding by the day. Here's the latest:

In the wake of Katherine Jackson losing contact with her grandkids - Paris, Prince and Blanket - TJ Jackson, the son of Michael's brother Tito, is pushing to become the children's temporary legal guardian. TMZ reports his attorney is actively drawing up the paperwork now.

Katherine has been serving as the trio's guardian since Michael Jackson passed away, but sources close to the family believe she's being held against her will by MJ's siblings in Arizona. She has not seen her grandchildren in nine days.

Insiders tell TMZ that all three children are in favor of TJ taking over guardian duties. He's reportedly very close to his niece and nephews.

"I know it's completely unfair for them to do this to you and your brothers. We will keep trying. I love you," TJ Tweeted to Paris last night.

Tension continues to rise within the Jackson clan because Michael only left money to his foundation and his children in his will. Moreover, there have been accusations that Katherine is being treated unfairly by the will's executors.

Police were called to the family home in California yesterday over some kind of domestic dispute. No arrests were made, but photos later surfaced of Janet and Jermaine Jackson in a heated discussion.

Finally, in case this wasn't all strange enough, there's this: TJ Jackson went to the prom with Kim Kardashian.

Syrian forces fight back after rebel surge into capital

BAB AL-HAWA, Syria/AMMAN (Reuters) - Syrian government forces pounded rebels in Damascus overnight, battling to reverse opposition gains in the aftermath of the assassination of President Bashar al-Assad's security chiefs.

Army helicopters and tanks aimed rockets, machineguns and mortars at pockets of rebel fighters who have infiltrated the capital this week in an operation they call "Damascus Volcano".

Lightly-armed fighters have been moving through the streets on foot and attacking security installations and roadblocks.

But the heart of the city was quiet by 0400 GMT on Saturday, residents told Reuters.

The Syrian Observatory for Human Rights, an opposition group which monitors the violence in the country, said 240 people had been killed across Syria on Friday, including 43 soldiers.

The Observatory's combined death toll over the past 48 hours stands at 550, making it the bloodiest two days of the 16-month-old uprising against Assad.

On Wednesday a bomb killed four members of the president's narrow circle of kin and lieutenants, including his powerful brother-in-law, defence minister and intelligence chief.

"The regime has been rudderless for last three days. But the aerial and ground bombardment on Damascus and its suburbs shows that it has not lost the striking force and that it is re-grouping," opposition activist Moaz al-Jahhar said by phone from Damascus.

In the days since, rebels have pushed deep into the heart of the capital and seized control of other towns. On Thursday, they captured three border crossings with Iraq and Turkey, the first time they have held sway over Syria's frontiers.

LOOTING

At Bab al-Hawa, a busy border post with Turkey seized by advancing fighters, rebels watched on with approval while jubilant villagers looted a duty free shop, part of the vast business empire of one of Assad's cousins.

"This is the people's money; they are taking it back," said rebel fighter Ismail. "Whoever wants to should take it."

Assad has failed to speak in public since Wednesday's blast. A funeral was held on Friday for three of the officials slain in the attack, but Assad did not attend and was nowhere to be seen.

A Damascus resident said he saw three tanks on the southern ring road late on Friday evening, firing at districts in west Damascus.

"The road was cut off and troops were firing mortar rounds from next to the tanks," he said.

A resident of Mezzeh, a district of high rise towers, villas and cactus fields, said helicopters were firing machineguns into the neighbourhood and rebels were firing back "uselessly" with automatic rifles.

A man in Barzeh, a neighbourhood to the northeast, said a barrage of mortar rounds began hitting residential buildings before midnight.

Loyalist snipers stationed in Ush al-Wawrar, an enclave in hills overlooking Barzeh populated mainly by members of Assad's Alawite minority sect, had killed a woman earlier in the day and there was gunfire between the two districts, he said.

Accounts could not be independently verified. The Syrian government restricts access by international journalists.

In at least one apparent success for Assad's forces, state TV said on Friday troops had cleared the central Damascus district of Midan of "mercenaries and terrorists". It showed dead men in t-shirts, some covered in blood, others burned.

Opposition activists and rebel sources confirmed they had withdrawn from that district after coming under heavy bombardment, but said they were advancing elsewhere.

"It is a tactical withdrawal. We are still in Damascus," Abu Omar, a rebel commander, said by telephone.

Assad's forces shelled the Abu Kamal crossing with Iraq on the Euphrates River highway, one of the most important trade routes in the Middle East, seized by rebels on Thursday.

A Reuters photographer at the scene said Iraqi forces had sealed off their side of the checkpoint with concrete walls. Late on Friday explosions and gunfire could be heard from the Syrian side, which had been burned and looted.

The surge in violence has trapped millions of Syrians, turned sections of the capital into ghost towns, and sent tens of thousands of refugees fleeing to neighbouring Lebanon.

END GAME?

Regional and world powers are now bracing for what could be the decisive phase of the conflict, hoping to wrench Assad out of power without unleashing a sectarian war that could spill across borders.

Israel said it would consider military action if needed to ensure Syrian missiles or chemical weapons did not reach Assad's allies in Lebanon, the Shi'ite Islamist movement Hezbollah.

"I have instructed the military to increase its intelligence preparations and prepare what is needed so that ... (if necessary) ... we will be able to consider carrying out an operation," Defence Minister Ehud Barak said.

Diplomacy has failed to keep pace with events. A day after Moscow and Beijing vetoed a U.N. resolution that would have allowed sanctions, the Security Council approved a 30-day extension of a small, unarmed observer mission, the only outside military presence on the ground.

"The regime is going through its last days," Abdelbasset Seida, the leader of the main Syrian opposition umbrella group, the Syrian National Council, said in Rome, predicting a dramatic escalation in violence.

(AP) ? An Air Force instructor implicated in a sweeping sex scandal at one of the nation's busiest military training bases was convicted in military court Friday of raping one female recruit and sexually assaulting several others.

Staff Sgt. Luis Walker, the first Lackland Air Force Base instructor to stand trial in the scandal, was found guilty by a jury of seven military personnel on all 28 counts he faced, including rape, aggravated sexual contact and multiple counts of aggravated sexual assault.

The jury deleted a clause from two counts that accused Walker of making flirtatious and lewd comments to trainees. However, it upheld the overall counts containing the deleted clauses, which accused him of trying to cultivate a sexual relationship with two trainees.

Walker faces up to life in prison and a dishonorable discharge at his sentencing hearing, which starts Saturday. He showed no emotion upon hearing the verdict. Outside the base courtroom afterward, Walker was met by his father and other relatives, some of whom were crying. He will remain free pending sentencing.

Walker is among 12 Lackland instructors investigated for sexual misconduct toward at least 31 female trainees. Six instructors have been charged on counts ranging from rape to adultery. Walker faced the most serious charges and was the first to stand trial.

Lackland is where every American airman receives basic training. It has about 475 instructors for the approximately 35,000 airmen who graduate every year. About one in five is female, pushed through eight weeks of basic training by a group of instructors, 90 percent of whom are men.

The sexual misconduct at the base apparently began in 2009, but the first woman didn't come forward until last year. The first allegations were levied against Walker, who is accused of crimes that allegedly took place between October 2010 and January 2011.

According to prosecutors, Walker had sexual intercourse with 4 of the 10 female recruits. He was also accused of making flirtatious or sexually suggestive comments, sending inappropriate text messages and sometimes groping his recruits.

Walker also is accused of forcing five recruits to engage in sexual acts by threatening their military careers and intimidating two of the women into lying about his alleged misconduct, prosecutors alleged.

Several of Walker's alleged victims testified during his court-martial, including one airman who described how Walker lured her into an office and sexually assaulted her on a bed, ignoring her pleas to stop.

The women told jurors that Walker gained their trust to get them alone in his office or an empty dormitory where he forced them into kissing, touching and intercourse. Those testifying said they didn't tell anybody at first because they feared being booted from the Air Force.

The Associated Press is not naming those who testified because they are alleged sexual assault victims.

Meanwhile, the case of another former Air Force training instructor has been referred to a general court-martial, according to an Air Force statement issued Friday evening.

Staff Sgt. Craig LeBlanc is charged with sexual misconduct, obstructing justice and making a false official statement. He is accused of using his post as a military instructor to sexually assault and pursue a sexual relationship with one female trainee, and have a wrongful sexual relationship with another. No trial date has been set.

One of the other instructors charged in the case, Staff Sgt. Peter Vega-Maldonado, pleaded guilty in June, admitting he had sex with a female trainee in exchange for a sentence of 90 days' confinement. He later acknowledged he had been involved with a total of 10 trainees ? a number previously unknown to investigators.

Islamist rebel group releases 3 hostages

BAMAKO, Mali (AP) ? Islamist rebels in north Mali on Wednesday released an Italian and two Spanish hostages who had been held since October, a spokesman with an allied group said.

The Movement for Unity and Jihad in West Africa, or MUJAO by its French acronym, released the hostages near the town of Gao, according to Sanda Abou Mohamed, a spokesman for an al-Qaida-linked rebel group Ansar Dine.

Spaniards Enric Gonyalons and Ainhoa Fernandez de Rincon and Italian Urru Scarlett are aid workers who were kidnapped from a refugee camp in southern Algeria in October, Mohamed said. He said that the Algeria-based MUJAO was responsible for their kidnapping, and in May requested money for their release. It is unclear if a ransom was paid.

Millions of dollars reportedly have been paid in ransoms for other hostages held by Islamic groups in the Sahara Desert.

At least six Western hostages were killed between 2009 and 2011. All were held by al-Qaida's African franchise, al-Qaida in the Islamic Maghreb or AQIM.

The Spanish Foreign Ministry confirmed the two Spaniards were being freed. It said the handover was "on the verge" of being completed but delayed at the last minute by a sandstorm.

A plane has been sent to Africa to pick them up, said a ministry official. He spoke on condition of anonymity in line with ministry rules.

Gonyalons worked for an NGO called Mundubat. Fernandez worked for the Association of Friends of the Saharawi People.

MUJAO is a breakaway from AQIM. Ansar Dine, led by a former Mali diplomat and one-time leader of a rebellion of the Tuareg people, is believed to have links to AQIM, which has claimed responsibility for numerous suicide bombings, as well as the kidnappings of at least 50 foreigners.

Northern Mali has become a magnet for Islamist radicals since Ansar Dine and AQIM fighters drove out separatist Tuareg rebels who had seized northern Mali in late March. The two Islamist groups want to impose Shariah law in the region.

This year Harry & David gave beautiful pear gift baskets to all of the EVO speakers, and we were able to participate in the ?Meet and Treat? where vendors who are a part of EVO have the opportunity to connect with other bloggers by hosting a giveaway, handing out coupons, or engaging in other creative interaction with attendees, during a sponsored breakfast.


So not only did we have a great time, with new friendships made, old ones rekindled, the power also went out one night (lots of excitement), and the weather was all over the map in this very beautiful setting. (The stormy view from our room.)

The mountains were as gorgeous as ever (you know how mountain weather goes, sunny one minute, rainy the next).

The closing party was a blast! (Me with Dine and Dish and I Wash You Dry.)

Good food, excitement, fun, friends ? you can?t ask for any more!

Ingredients

  • 1 10-oz. bag of Harry and David dark chocolate truffles
  • 1 10-oz. bag of Harry and David Moose Munch snack
  • 4 large bananas

Instructions

  1. Place truffles in a microwave-safe container. Microwave at 50% power for 1-minute at a time. Stir in-between intervals, for a total of approximately 3 minutes, until melted.
  2. Place Moose Munch snack into a large resealable bag; use a rolling pin to crush into small pieces.
  3. Peel and cut the bananas into approximately 2-inch lengths, then dip each piece of banana into the melted chocolate and roll in the crushed Moose Munch snack.
  4. Place on baking sheet and refrigerate for 15-minutes before serving.

New insights into how the most iconic reaction in organic chemistry really works

ScienceDaily (July 9, 2012) ? In 1928, chemists Otto Diels and Kurt Alder first documented diene synthesis, a chemical reaction important for synthesizing many polymers, alkaloids and steroids. Their work on this mechanism, which came to be known as the Diels-Alder reaction, won them the 1950 Nobel Prize in chemistry.

Since then, the iconic reaction has become the most commonly used and studied mechanism in organic chemistry. But what happens during the reaction has never been entirely clear.

Now, Kendall N. Houk, UCLA's Saul Winstein Professor of Organic Chemistry, and colleagues report exactly how the Diels-Alder reaction occurs. Their research is published this week in the early online edition of the journal Proceedings of the National Academy of Sciences and will be published in an upcoming print edition.

"We have examined the molecular dynamics of the Diels-Alder reaction, which has become the most important reaction in synthesis, in detail to understand how it happens," said Houk, who is a member of the California NanoSystems Institute at UCLA.

Houk and his colleagues created a number of simulations -- he calls them short movies -- of molecules coming together and reacting.

"The idea," Houk said, "is to understand how the reaction happens -- not just that A goes to B and B goes to C, but to actually follow how the bonds are forming and how the atoms are moving as these things come together. Using the massive computing power we have now, we get a degree of resolution of the mechanism that was not really possible before. It took a lot of computer time, but as a result, we now have unprecedented insight into how this reaction occurs."

Organic chemists have argued about this for years: If two bonds form during a reaction, do they form at the same time, or does one form first and then the other?

"We find that for the simplest Diels-Alder cycloaddition, it takes only about five femtoseconds on average between the formation of the two bonds; we consider that as occurring simultaneously," Houk said. (A femtosecond is approximately one millionth of one billionth of a second.)

Houk's new PNAS paper is his first in the journal since being elected to the National Academy of Sciences in 2010. The same PNAS issue also features an interview with Houk, who is one of the most prolific chemists in the world and one of the world's leading physical organic chemists.

"We have studied many different classes of reactions and come up with various kinds of rules for understanding why things happen the way they do," Houk said in the interview.

He and his colleagues -- who include David Baker at the University of Washington, Charles Doubleday at Columbia University and Kersey Black at Claremont McKenna College -- use computational methods to better understand basic chemical reactions and to design proteins and enzymes to catalyze chemical reactions. The combination of computational design and molecular biology "leads to a catalyst for whatever reaction is needed, if we can get this all to work properly," Houk said.

Describing his research to predict the structure of novel proteins that could catalyze specific chemical reactions, he said, "The idea is to design a catalyst for any reaction that's important for whatever reason -- an important drug or a commercial product, for example."

Designing proteins and enzymes is difficult, but it can be successful.

"We've beaten really enormous odds but have really just scratched the surface of what is possible," Houk said. "Future developments in theory and computing power should make this work much better."

Black and Doubleday are co-authors with Houk on the new PNAS article, along with Peng Liu, a postdoctoral scholar in Houk's laboratory, and Lai Xu, who earned her Ph.D. in Houk's lab and is currently a postdoctoral scholar at Texas Tech University. (Liu, who also conducted his Ph.D. research in Houk's lab, was awarded a UCLA Chancellor's postdoctoral award last month.)

Houk has pioneered the use of computer calculations and simulations to study organic chemistry and to predict chemical reactivity. His research group has made predictions of new phenomena that have been verified experimentally, and he has made critical contributions to our understanding of how enzymes are able to selectively catalyze reactions.

In 2008, his research group used computer methods to create "designer enzymes" and to predict structures of proteins that can catalyze reactions which do not occur naturally -- a major milestone in computational chemistry and protein engineering. Designer enzymes are likely to have applications for defense against biological warfare by deactivating pathogenic biological agents, and for creating more effective medications, Houk said.

He and his colleagues are currently working on computational methods to predict catalysts for reactions that will have important applications in industry and in therapies for fighting disease.

"Our work is theoretical and computational but is always tied to real phenomena," Houk said. "We first try to understand what is happening and then try to make predictions that experimentalists can test. The goal of our research is to use computational methods to design the arrangement of groups inside a protein to cause any desired reaction to occur."
