Viruses and mucus team up to ward off bacteria

Phages may play unforeseen role in immune protection

The last thing most people would want in their bodies is mucus laden with viruses. But a new study suggests that viruses called bacteriophages, or phages, grab onto mucus and then infect and kill invasive bacteria. The finding, reported May 20 in the Proceedings of the National Academy of Sciences by Forest Rohwer of San Diego State University and colleagues, could mean that some viruses partner with animals and humans to stave off bacterial infections and control the composition of friendly microbes in the body.

Bacteriophages are viruses that break open bacteria, killing them. Researchers have studied bacteriophages for decades, and some disease therapies take advantage of the viruses? bacteria-slaying abilities, says microbiologist Frederic Bushman of the University of Pennsylvania medical school. But the study provides what Bushman says is a revelation that should have been obvious; phage may be a natural part of the immune system. ?It?s new in a way that is sort of common-sensey,? he says.

Previously, researchers thought of mucus mainly as a physical barrier to keep invading organisms from entering the body. The slimy substance made by our noses, intestines and other organs also fights invaders with antimicrobial molecules. ?Some researchers had found bacteriophages stuck in mucus, but they figured that the mucus had stopped or slowed the viruses. No one realized that the viruses are part of the body?s defense, says study coauthor Jeremy Barr, who works in Rohwer?s lab. ?This is a natural use of phage therapy that has probably been around since mucosal surfaces evolved,? Barr says.

Rohwer, who studies corals, had noticed that phages tend to concentrate in mucus. To find out why, the researchers collected mucus from human gums, sea anemones, fish, corals and mouse intestines. Mucus layers had more phages and fewer bacteria than the surrounding environment, suggesting that the viruses helped to limit the number of bacteria allowed into the mucus.

Phages are coated in proteins that latch onto sugars called glycans, anchoring the viruses in the mucus, the team discovered. From there the phages can ambush encroaching bacteria.

So far, the researchers have demonstrated that mucus and phages can work together to protect cells in a dish. The next step, Bushman says, would be determining what happens inside an organism, an experiment the researchers are already planning.

Asia stocks mixed as holidays thin trade

(AP) ? Asian stock markets were mixed in holiday-thinned trading Friday as investors digested a slew of disappointing economic data and corporate results from the U.S.

Applications for unemployment benefits jumped to their highest level in six weeks, the U.S. Labor Department reported Thursday, while manufacturing slowed in the mid-Atlantic region.

The bright spot was applications for new construction, which reached a five-year peak, reinforcing "the patchy nature of the US economic recovery," Michael Hewson of CMC Markets said in a commentary.

Japan's Nikkei 225 index fell 0.1 percent to 15,019.55. Australia's S&P/ASX 200 rose 0.4 percent to 5,186.90. Benchmark in mainland China and Indonesia rose while those in New Zealand and the Philippines fell. Markets in Hong Kong and South Korea were closed for public holidays.

Corporate earnings, which have been helping to power Wall Street to all-time highs, took a step back on Thursday after Wal-Mart, the world's largest retailer, reported a disappointing first-quarter profit and acknowledged a sales slump. Personal computer maker Dell posted dismal first quarter earnings.

The Dow Jones industrial average fell to 15,233.22, a loss of 0.3 percent. The Standard & Poor's 500 dropped 0.5 percent to 1,650.47. Both indexes closed at record highs the day before. The Nasdaq composite index fell 0.2 percent to 3,465.24.

Benchmark oil for June delivery was down 16 cents to $95 per barrel in electronic trading on the New York Mercantile Exchange. The contract rose 86 cents to $95.16 per barrel on the Nymex on Thursday.

In currencies, the euro fell to $1.2862 from $1.2907 late Thursday in New York. The dollar rose to 102.28 yen from 102.06 yen.

Nichols has a strong feeling for the tactility of natural elements-water, wood, terrain, weather.

Nichols takes his time with the story, dwelling on how the boy is shaped by the killer's tragic sense of romance, yet the suspense holds.

"Mud" isn't just a movie. It's the firm confirmation of a career.

"Mud" unfolds at its own pace, revealing its story in slivers. The performances are outstanding, especially from Sheridan, who plays tough, sweet, vulnerable and confused with equal conviction.

The film is drenched in the humidity and salty air of a Delta summer, often recalling the musical, aphoristic cadences of Sam Shepard, who happens to appear in a supporting role.

A wonderful, piquant modern-day variation on "Huckleberry Finn.''

Nichols is a gifted writer-director who knows how to get into the heads of his characters. And this film has superior actors who create people who are intriguing and hugely involving.

'Mud' is a standout film in this 'coming of age' genre mainly because of its central character, one tough, warm-hearted, stubborn little kid who believes in the power of love, above all else.

Other than pacing problems that needlessly stretch the film past the two-hour mark, 'Mud' slings the dirt and sweat with the best of them, as it both mourns and celebrates a way of life that's all but disappeared.

A sublime coming-of-age film, 'Mud' would be the offspring if 'Stand By Me' and 'Cape Fear' reproduced.

Mud is a movie of striking performances and memorable images and of people who seem to belong in rather than being imposed upon their environment.

A brilliant metaphor for how a child deals with divorce.

This might be Mud. But it deserves to stick.

This is a film that gives McConaughey the chance to be an actor rather than a star, and, not for the first time, he grabs the opportunity.

Gone is the rom-com character and emphasis on a toothy smile and six-pack abs. Matthew McConaughey is showing off some acting chops.

Mud is, perhaps, a little longer than it needed to be, but few sensitive viewers will begrudge Nichols his indulgence. This director is the real thing.

Writer-director Nichols continues to get inside the heads of his characters with this involving but overlong dramatic thriller.

McConaughey is terrific but the picture is really made by the fierce and unforgettable performance of Tye Sheridan as Ellis.

a film that demands a certain patience, which it rewards with various emotional payoffs that aren't ever quite what you expect

Somewhere in the dreams of Mud creator Jeff Nichols, Huckleberry Finn met Aguirre, Wrath of God.

Impressively directed and beautifully shot, this is an emotionally engaging and powerfully evocative coming-of-age drama with a superb script and terrific performances from a note-perfect cast.

This richly rewarding work lightly wears influences as disparate as Huckleberry Finn and The Hustler but ultimately emerges as the unmistakeable work of Nichols, one of the rare breed of genuinely exciting directors working in Hollywood today.

The disappointing denouement is far outweighed by the overall strength of the journey to that resolution.

Nichols keeps getting better, but his masterpiece is (excitingly) still to come.

Matthew McConaughey is as compelling as we've come to expect since The Lincoln Lawyer kicked off his 2010s hot streak, as Jeff Nichols ably combines elements of Terence Malick with an all-American rural yarn.

Mud clearly sets out from frame one to run along well-worn tracks - it's like Tom Sawyer and Huck Finn meets Whistle down the Wind.

Vintage Target Poster Similar To Mad Men Ad - Business Insider

Illustrator Brian Sanders' hand-drawn, vintage-style poster for the "Mad Men" season 6 premiere drove fans wild with its hidden plot clues.

It drove Target historian ? yes, that's a real thing?? Tony Jahn crazy for another reason: He felt like he'd seen the ad before.

It turns out that the AMC commissioned poster bears a striking resemblance to a 1960s ad campaign from Dayton, Target's predecessor.

Federal Judge: Only Powered-Off Cell Phones Deserve Privacy Protections

A federal magistrate judge in New York recently ruled that cell phone location data deserves no protection under the Fourth Amendment and that accordingly, the government can engage in real-time location surveillance without a search warrant. In an opinion straight from the Twilight Zone, magistrate judge Gary Brown ruled two weeks ago that ?cell phone users who fail to turn off their cell phones do not exhibit an expectation of privacy.?

The case in question involved a physician who the DEA believed had issued thousands of prescriptions for pain killers in exchange for cash. In March of this year, the DEA had obtained a warrant for his arrest, and, not knowing where he was, sought an order from magistrate judge Brown forcing the phone company to provide real-time data identifying the location of the physician?s phone.

Although the DEA agents requested a search warrant and the judge found that there was probable cause to believe that the cell phone location data would assist in the location and apprehension of an individual for whom there was already a valid arrest warrant, the judge later published a 30-page opinion further stating that he didn?t think the government needed to seek a search warrant in the first place.

Don?t Want the Government Tracking You? Turn Your Phone Off

In his puzzling opinion, the judge squarely criticizes people naive enough to expect privacy while also leaving their cell phones on when they?re not using them.

Given the ubiquity and celebrity of geolocation technologies, an individual has no legitimate expectation of privacy in the prospective location of a cellular telephone where that individual has failed to protect his privacy by taking the simple expedient of powering it off.

?

As to control by the user, all of the known tracking technologies may be defeated by merely turning off the phone. Indeed?excluding apathy or inattention?the only reason that users leave cell phones turned on is so that the device can be located to receive calls. Conversely, individuals who do not want to be disturbed by unwanted telephone calls at a particular time or place simply turn their phones off, knowing that they cannot be located.

The Catch-22 here is that the only people who the judge believes would have any reasonable expectation of privacy are those whose phones are turned off (and thus, not generating any location data that the government could access, even with a warrant). And it ignores the necessity of keeping your cell phone turned on for communicating with family or for work.

That consumers are dumb enough to willingly share their location using the ?Girls Around You? app (which the judge specifically calls out by name), only further justifies covert, warrantless government surveillance:

Given the notoriety surrounding the disclosure of geolocation data to retailers purveying soap powder and blue jeans to mall shoppers, the police searching for David Pogue?s iPhone and, most alarmingly, the creators and users of the Girls Around You app, cell phone users cannot realistically entertain the notion that such information would (or should) be withheld from federal law enforcement agents searching for a fugitive.

This is, in a word, ridiculous. There is a big difference between location information you knowingly share with a select group of friends (or, in fact, the world) and information collected about you without your knowledge or consent. Someone might be happy to share their location with a few friends by ?checking in? using Foursquare while at a music festival, but not want law enforcement to access that same information. And, they would still reasonably expect that their location a week later while at an Alcoholics Anonymous meeting or abortion clinic should remain private. Sharing location data isn?t and shouldn?t be all or nothing.

We are also baffled by the judge?s willingness to tie a reasonable expectation of privacy to the use of a cellphone power button. We?re not sure if the judge has watched the Onion?s spoof news video describing a fictional ?Google Opt Out Village? for people who don?t want to be tracked by the advertising company, but the logic in his opinion is consistent with the absurdity of that spoof. If you don?t want Google to track you, stop using all modern technology and move to a remote village. If you don?t want the government to covertly track your phone, turn it off and leave it off. What could be simpler, right?

Prince Harry end U.S. visit with polo match

GREENWICH, Conn. (AP) ? Britain's Prince Harry helped lead a polo team to victory in a charity match before a society crowd Wednesday in Greenwich, wrapping up a weeklong visit to the United States in which he also toured areas devastated by Superstorm Sandy and avoided the kind of salacious headlines stirred by his last American tour.

The prince scored a game-tying goal in the match to benefit Sentebale, the charity he co-founded to help poor children and AIDS orphans in the small African nation of Lesotho.

"Thank you for a wonderful week," Prince Harry said in a speech before taking to the manicured field at the Greenwich Polo Club. "I have witnessed the extraordinary generosity of the people of this great nation."

The match drew an invitation-only crowd of about 400 including supermodel Karolina Kurkova and fashion designers Jason Wu and Valentino, who posed for pictures on a red carpet at the club on their way to lunch with the prince.

"Is the sun coming out?" Kurkova asked as she posed in a light rain.

Greenwich, a New York City suburb, ranks among the wealthiest towns in America and is hardly unaccustomed to fanfare. But for some, the royal visit of the 28-year-old prince still was cause for celebration.

At the Atelier360 shop on Greenwich Avenue, the company was holding a party with tea and cucumber sandwiches. Co-owner Veronique Lee said British designers are well represented at the boutique, which sells items including hand bags, flasks and cuff links.

"We like royalty. What can you say?" Lee said. "We're having a lot of fun here."

The polo club, founded by billionaire Peter Brant in 1981, has hosted other British royals including the Duke and Duchess of York, Prince Andrew and Sarah Ferguson, in 1987 and Torquhil Campbell, the Scottish Duke of Argyll, in 2012. Brant said Wednesday that Harry's father and grandfather were good players and he was excited to watch Harry play at his club.

"It's a great honor," he said "It's great for polo."

Dressed in a navy-colored suit, white shirt and no tie, the prince arrived at the club in the late morning and was greeted by Brant and his wife, model Stephanie Seymour. They viewed a selection of art from the Brant Foundation Art Study Center as well as some paintings owned Brant, including some pieces by Andy Warhol, in the club's entrance hall.

After trying his hand at baseball in New York City on Tuesday, Harry showed his athleticism playing "the sport of kings." Harry scored a goal to tie the polo match at 3-3, and his team went on to a 4-3 win. Dawn Jones, the wife of actor Tommy Lee Jones, scored for the opposing team and was named most valuable player.

Connecticut Gov. Dannel Malloy was not among the invited. "He's more of a hockey guy, anyway," Malloy spokesman David Bednarz told the Greenwich Time.

Prince Harry also touted the work of Sentebale and thanked people for their support of the charity. Sentebale ? which means "forget-me-not" ? is a charity founded by Harry and Lesotho's Prince Seeiso that helps children struggling with poverty in the tiny southern African country.

"The HIV pandemic continues to leave thousands of children without parents and family structures to guide them through life," the prince said. "Without this support, basic needs such as food, shelter and care remain unmet, leaving children vulnerable and very often without much hope in their lives."

On his last U.S. visit, the third-in-line to the British throne stormed into the headlines last year when he was caught frolicking in the nude with a woman after an alleged game of strip billiards in his Las Vegas hotel room.

The Greenwich visit struck a lighter tone than his stop the previous day in New Jersey, where he toured two shore communities devastated by Superstorm Sandy.

New Jersey Gov. Chris Christie showed the prince a spot in Mantoloking where the sea had cut the town in half, taking out a bridge and houses. The channel has since been filled in. Every one of the wealthy town's 521 homes was damaged or destroyed. Scores remain as piles of rubble.

"This used to be a house?" Prince Harry asked at one barren spot.

The prince said he was impressed to see "everyone getting together and making things right."

He also spent Tuesday in New York City at events promoting tourism, entrepreneurism and philanthropy.

The prince, the son of Prince Charles and the late Princess Diana, began his visit to the U.S. on May 9. He is third in line to the British throne, after his father and older brother, Prince William.
